Performance Evaluation of the Automated Urine Microscopic Analyzer UF5000i Using Imaging and Flow cytometric method

Abstract
Purpose : Automated urine microscopic analyzers are very useful in the clinical laboratory that examine large quantities of samples because they can report faster and objectively, and also require less labor. We evaluated the performance of the automated urine microscopic analyzer UF5000i.

Materials and Methods : The within-run and between day precisions of RBC, WBC, epithelial cells and bacteria were measured using two levels of control materials. The agreement of UF5000i and manual microscopy or UF1000i was measured. The linearity, recovery rate and review rate of UF5000i were evaluated.

Results : The linearity for RBC and WBC was good (R2>0.999) and recovery range for RBC, WBC and bacteria was 98.5%-102.2%. The carry over was not observed in these items. Within run precision for RBC, WBC, epithelial cells and bacteria were 6.9%, 4.9%, 13.0%, 7.1% in low concentration, and 2.5%, 1.9%, 12.7%, 3.4% in high concentration, respectively. Between run precision for RBC, WBC, epithelial cells and bacteria were 5.9%, 5.0%, 23.8%, 6.5% in low concentrations and 4.0%, 4.0%, 16.9%, 4.5% in high concentration, respectively. The agreements with manual microscopy in RBC and WBC were 96.9% and 97.9%. The agreements with UF1000i in RBC, WBC and bacteria were 97.7%, 97.7%, 99.1% and 95.9%, respectively. The review message of UF5000i(12.2%) was lower than UF1000i(26.1%).

Conclusion: The precision, linearity and agreement of UF5000i were excellent in this study. Therefore, this device will be useful as a screening analyzer of urine microscopic test.
